the biggest example i can think of is The Walkmen's "The Rat"...the song was so dominantly overwhelmingly popular, fans lost interest in all the other songs, whether new or old, and the band grew sick of the song, because it's the only one the fans wanted to hear, band members started talking about "out of body experience" while playing the song, because they were so sick of it and so used to playing it over and over

Those were fantastic, but not defining songs, as those artists had many, many hit. Defining songs are often defining for one hit wonders or artists with a small repertoire. Such as:
