I am sure a few would be opposed on the basis of their religious beliefs but admittedly I have zero idea how high or low the number. Either way, it is tough to judge b/c at this point we are discussing a hypothetical.
But that's the thing, we are not. He is working on it NOW. And they are objecting. Its not going to go anywhere BECAUSE of the very base that elected him.

So now you have two long standing roots of your base pitted AGAINST each other by your own party INSIDE the party. THAT is a problem.

It is not going to go anywhere b/c the GOP controlled Congress is never going to allow the bill to pass. Besides, I am not seeing this wide spread opposition to adding gays as a protected class to The Civil Rights Act from African Americans elected to Congress. Hell, John Lewis is co-sponsoring the bill. It seems the concerns from civil rights group are over poison-pill amendments or using the debate to expanded religious accommodations in even further.
